Acid-base properties for ruthenium(II) bis(2,2'-bipyridine) 3-carboxyl-2,2'-bipyridine reveal a ground state pK(a) of 0.82 +/- 0.07 and an excited state pK(a) of 2.31 +/- 0.05, a 1.5 pH unit increase from the ground state. The excited state pK(a) is temperature independent while the ground state pK(a)(0) increases with temperature and has DeltaH(0) and DeltaS(0) values of -990 +/- 149 cm(-)(1) and -4.57 +/- 0.48 cm(-)(1) K(-)(1), respectively. The acidic form of the complex emits at lower energy than the basic form at both 296 and 77 K. The emission energy maxima are solvent dependent and decrease in energy when the solvent changes from 4:1 (v/v) 2-MeTHF-CH(2)Cl(2) to water and when the pH decreases. Changes in excited state lifetimes with emission energy follow the energy gap law with an intercept of 49 +/- 1 and a slope of (2.11 +/- 0.09) x 10(-)(3). Emission quantum yields for protonated and deprotonated species in 4:1 (v/v) 2-MeTHF-CH(2)Cl(2) are 0.023 +/- 0.001 and 0.110 +/- 0.002, respectively. The temperature dependence of the emission lifetimes gives energy barriers of 270 cm(-)(1) for the complex in aqueous solution at pH -0.5, and 990 cm(-)(1) in aqueous solution at pH 4.5, and 1920 cm(-)(1) in 4:1 (v/v) 2-MeTHF-CH(2)Cl(2.)  相似文献

Temperature-responsive random copolymers based on poly(N-isopropyl acrylamide) (PNIPAM) with 15 mol% of either acrylic acid or dimethylaminoethyl acrylate quaternary chloride were prepared. The effect of the charge and its sign were investigated in the solid-liquid separation of silica and alumina mineral suspensions. The results were compared to PNIPAM homopolymer of similar molecular weight. PNIPAM copolymers of the same charge as the particles (co-ionic PNIPAM) act as dispersants at both 25°C and 50°C. Flocculation occurs when counter-ionic PNIPAM facilitates selective aggregation and rapid sedimentation of minerals at both 25°C and 50°C. Adsorption and desorption studies showed that, unlike non-ionic PNIPAM, little desorption of the counter-ionic copolymers from the oxides occurred after cooling a suspension from 50°C to below the lower critical solution temperature. Thus, incorporation of counter-ionic charge into the temperature sensitive polymer PNIPAM was found to reduce the sediment bed consolidation upon cooling when compared to PNIPAM homopolymers. The lack of secondary consolidation upon cooling is attributed to attractive inter-particle forces, such as conventional polyelectrolyte flocculation mechanisms (bridging, charge neutralization or charge patch) which persist at both 25°C and 50°C when counter-ionic PNIPAM is used. On the other hand, it was possible to obtain rapid sedimentation with the counter-ionic PNIAPMs even when they were added to the suspension already at 50°C, a process which has not been possible with neutral PNIPAM homopolymers.  相似文献

The design, construction and operation of a simple, inexpensive and compact high voltage power supply (HVPS) for use in conjunction with a simple cross capillary electrophoresis microchip is presented. The microchip HVPS utilizes a single high voltage power supply (15 kV), a voltage-divider network, to give the voltages necessary to operate a gated injection valve, and two high voltage relays for switching between the open and closed gate sequences of the injection. In order to accommodate the application of different simple cross microchip dimensions, a set of equations for defining the resistor network and ensuring proper gate performance are presented.  相似文献

A novel PLA-based polymer containing reactive pendent ketone or hydroxyl groups was synthesized by the copolymerization of L-lactide with epsilon-caprolactone-based monomers. The polymer was activated with NPC, resulting in an amine-reactive polymer which was then cast into thin polymeric films, either alone or as part of a blend with PLGA, before immersion into a solution of the cell adhesion peptide GRGDS in PBS buffer allowed for conjugation of GRGDS to the film surfaces. Subsequent 3T3 fibroblast cell adhesion studies demonstrated an increase in cellular adhesion and spreading over films cast from unmodified PLGA. Hence the new polymer can be used to obtain covalent linkage of amine-containing molecules to polymer surfaces.  相似文献

Geldanamycin, a polyketide natural product, is of significant interest for development of new anticancer drugs that target the protein chaperone Hsp90. While the chemically reactive groups of geldanamycin have been exploited to make a number of synthetic analogs, including 17-allylamino-17-demethoxy geldanamycin (17-AAG), currently in clinical evaluation, the "inert" groups of the molecule remain unexplored for structure-activity relationships. We have used genetic engineering of the geldanamycin polyketide synthase (GdmPKS) gene cluster in Streptomyces hygroscopicus to modify geldanamycin at such positions. Substitutions of acyltransferase domains were made in six of the seven GdmPKS modules. Four of these led to production of 2-desmethyl, 6-desmethoxy, 8-desmethyl, and 14-desmethyl derivatives, including one analog with a four-fold enhanced affinity for Hsp90. The genetic tools developed for geldanamycin gene manipulation will be useful for engineering additional analogs that aid the development of this chemotherapeutic agent.  相似文献

Four isoforms of the extracellular lignin peroxidase of the ligninolytic actinomyceteStreptomyces viridosporus T7A (ALip-P1, P2, P3, and P4) were individually purified by ultrafiltration and ammonium sulfate precipitation, followed by electro-elution using polyacrylamide gel electrophoresis. Three of the purified peroxidases were compared for their immunologic relatedness by Western blot analysis using a polyclonal antibody preparation produced in rabbits against pure isoform P3. The anti-P3 antibody was also tested for its reactivity towards a lignin peroxidase from the white-rot fungusPhanerochaete chrysosporium and another ligninolytic actinomyceteStreptomyces badius 252. Results showed that peroxidases ALip-P1 through ALip-P3 are immunologically related to one another. The peroxidases ofS. badius, but not the peroxidase ofP. chrysosporium, also reacted with the antibody, thus indicating that the lignin peroxidases ofS. viridosporus andS. badius are immunologically related. Based upon its specific affinity, lignin peroxidase isoform ALip-P3 ofS. viridosporus was readily purified using an anti-P3 antibody affinity column.  相似文献

We present data on the coverage and nearest-neighbor dependences of the diffusion of CO on Cu(111) by time-lapsed scanning tunneling microscope (STM) imaging. Most notable is a maximum in diffusivity of CO at a local coverage of one molecule per 20 substrate atoms and a repulsion between CO molecules upon approach closer than three adsites, which in combination with a less pronounced increase in potential energy at the diffusion transition state, leads to rapid diffusion of CO molecules around one another. We propose a new method of evaluating STM-based diffusion data that provides all parameters necessary for the modeling of the dynamics of an adsorbate population.  相似文献

Several procedures are available for simulating and optimising separations in ion chromatography (IC), based on the application of retention models to an extensive database of analyte retention times on a wide range of columns. These procedures are subject to errors arising from batch-to-batch variability in the synthesis of stationary phases, or when using a column having a different diameter to that used when the database was acquired originally. Approaches are described in which the retention database can be recalibrated to accommodate changes in the stationary phase (ion-exchange selectivity coefficient and ion-exchange capacity) or in the column diameter which lead to changes in phase ratio. The entire database can be recalibrated for all analytes on a particular column by performing three isocratic separations with two analyte ions. The retention data so obtained are then used to derive a "porting" equation which is employed to generate the required simulated separation. Accurate prediction of retention times is demonstrated for both anions and cations on 2mm and 0.4mm diameter columns under elution conditions which consist of up to five sequential isocratic or linear gradient elution steps. The proposed approach gives average errors in retention time prediction of less than 3% and the correlation coefficient was 0.9849 between predicted and observed retention times for 344 data points comprising 33 anionic or cationic analytes, 5 column internal diameters and 8 complex elution profiles.  相似文献

Cosmetic products that contain retinyl palmitate are popular as antiaging skin treatments; however, recent studies suggest a risk for enhanced skin tumor development with topical retinyl palmitate applications and exposure to solar ultraviolet radiation (UVR). In this study, we investigated the potential of retinyl palmitate to enhance UVR‐induced photo‐co‐carcinogenesis. Groups of 36 male and 36 female SKH‐1 hairless mice were exposed to simulated solar light (SSL) and treated with the control cream or creams containing retinyl palmitate, 5 days per week for 40 weeks. Other groups of mice were exposed to SSL and received no cream treatment or received cream treatments and were exposed to ultraviolet‐A or ultraviolet‐B. Mice were monitored for the development of skin tumors, and the incidences and multiplicities of squamous cell neoplasia were determined by histopathology. In both the absence and presence of SSL, mice administered the control cream developed skin tumors earlier and had higher incidences and multiplicities of skin squamous cell neoplasms than mice that received no cream treatment. Compared to the control cream groups, mice exposed to SSL and administered the retinyl palmitate creams demonstrated earlier onsets of skin tumors and had increased incidences and multiplicities of squamous cell skin neoplasms.  相似文献
